目的:研究灵芝糖萜复合微乳的制备工艺,同时考察其理化性质及其对移植性肝癌Heps小鼠的抗肿瘤作用。方法:采用伪三元相图得到空白微乳的处方,进而制备灵芝糖萜复合微乳;采用透射电镜及激光粒度分析仪考察糖萜微乳的形态、粒径分布和Zeta电位;测定微乳对实体瘤小鼠的抑瘤率,并比较各组实体瘤小鼠的体重、免疫器官(脾和胸腺)指数。结果:灵芝糖萜O/W型微乳的最佳处方为聚氧乙烯脱水山梨醇月桂酸酯(吐温20)-异丙醇-水-油酸体系,其比例为14.3∶14.3∶33.3∶2;透射电镜观察乳滴呈圆球形,大小均匀;平均粒径32.43 nm,Zeta电位-3.41 mV;糖萜微乳低高剂量组(57.25,114.5mg.kg-1.d-1)的抑瘤率分别为37.66%,52.34%。结论:灵芝糖萜微乳粒径小,稳定性好,对小鼠移植性肝癌Heps有显著的抑制作用。
OBJECTIVE: To study the preparation technology of Ganoderma lucidum saccharide ternary composite microemulsion, investigate its physicochemical properties and anti-tumor effects on transplanted liver cancer Heps mice. Methods: The formulation of blank microemulsion was obtained by using pseudo-ternary phase diagram to prepare Ganoderma lucidum glycoside composite microemulsion. The morphology, particle size distribution and Zeta potential of the terpene microemulsion were investigated by transmission electron microscopy and laser particle size analyzer. The tumor inhibition rate of the solid tumor mice was compared with that of the solid tumor mice in each group. The indexes of immune organs (spleen and thymus) were compared. Results: The best prescription of Ganoderma lucidum O / W microemulsion was polyoxyethylene sorbitan laurate (Tween 20) -isopropanol-water-oleic acid system with the ratio of 14.3:14.3:33.3: 2; The transmission electron microscopy showed the spherical droplets were spherical, uniform size; average particle size of 32.43 nm, Zeta potential -3.41 mV; low-dose group of terpene microemulsion 57.25,114.5mg.kg-1.d-1 The tumor rates were 37.66% and 52.34% respectively. CONCLUSION: Ganoderma lucidum saccharide ternary microemulsion has small particle size and good stability, and it has significant inhibitory effect on Heps of transplanted hepatocarcinoma in mice.
