目的 探讨 PTEN和组织蛋白酶 - B(CB)表达与星形细胞瘤恶性程度和侵袭性的关系 .方法 应用免疫组化方法检测 5 6例星形细胞瘤标本中 PTEN和 CB的表达 ,分析其与肿瘤恶性程度和侵袭性的关系 .结果 PTEN阳性染色主要定位于细胞质中 ,5 6例星形细胞瘤中 PTEN阳性表达 34例 (6 1% ) ,高分化组 ( 和 级 ) 2 3/2 7例 (85 % )和低分化组( 和 级 ) 11/2 9例 (38% )之间差异显著 ,明显高于低分化组 (P<0 .0 1) .CB阳性染色主要定位于细胞质中 ,呈棕黄色细颗粒状 .在高分化组中 ,呈散在的弱阳性表达 ,表达率为37% (10 /2 7) ;而在低分化组 ,表达明显增强 ,且侵袭前沿和瘤周 ,表达率为 6 6 % (19/2 9) ,与低分化组有显著差异 (P<0 .0 5 ) .PTEN和 CB共同表达者 8例 ,其中低分化组 1例 ,在高分化组中 7例 ,两者相比差异有显著性意义 (P<0 .0 5 ) .结论 PTEN基因突变或缺失在星形细胞瘤的发生发展中起重要作用 ,与肿瘤恶性分化程度密切相关 ,PTEN缺失与侵袭性有关 ,是低分化肿瘤侵袭性高的原因之一
Objective To investigate the relationship between the expression of PTEN and cathepsin-B (CB) and the malignancy and invasiveness of astrocytomas.Methods The expressions of PTEN and CB in 56 specimens of astrocytoma were detected by immunohistochemistry, Tumor grade and invasiveness.Results Positive staining of PTEN mainly located in the cytoplasm, positive expression of PTEN was found in 34 cases (61%) of 56 astrocytomas, high grade (grade 2/3 7) There was a significant difference between cases (85%) and 11/2 (38%) in poorly differentiated group (vs grade), which was significantly higher than that in poorly differentiated group (P <0.01) .CB positive staining mainly localized in cytoplasm , Showed a yellowish brown granular.The weakly positive expression was scattered in the well-differentiated group, the expression rate was 37% (10/27), while in poorly differentiated group, the expression was significantly enhanced, and the invasion front and tumor, The expression rate was 66% (19/2 9), which was significantly different from that in poorly differentiated group (P <0.05) .8 cases were co-expressed with PTEN and CB, of which 1 was poorly differentiated 7 cases, the difference was significant (P <0.05) .Conclusion PTEN gene mutation or deletion plays an important role in the development of astrocytoma Role, is closely related to the degree of differentiation of malignant tumors, PTEN deletion and aggressiveness, is one of the reasons poorly differentiated high tumor aggressiveness
