研究载脂蛋白 (Apo)E基因多态性在吉林地区正常汉族人群中的分布状态。方法 :应用聚合酶链反应———限制性片段长度多态性 (PCR -RFLP)方法先扩增出ApoE基因第 4外显子内的 2 2 7bp片段 ,继以限制性切酶Cfol酶解消化 ,8%聚丙烯酰胺凝胶电泳 ,最后银染显带判定基因型 ,并计算等位基因频率。结果 :本实验共检测分析了 2 0 6例吉林地区正常汉族人的ApoE基因型及等位基因频率。结果表明纯合子ε3 /3 基因型频率及ε3 等位基因频率最高。ε2 高于ε4。且ε2 随年龄增长有增加趋势。结论 :该方法快速、准确 ,分辨率高。ApoE基因多态性分布的研究为ApoE生理功能的研究及进一步临床应用打下基础。
To investigate the distribution of apolipoprotein (Apo) E gene polymorphism in normal Han population of Jilin province. Methods: The PCR-RFLP method was used to amplify a 227 bp fragment of exon 4 of ApoE gene and then cleaved with restriction enzyme Cfol , 8% polyacrylamide gel electrophoresis, the last silver stain band to determine the genotype, and calculate the allele frequency. Results: ApoE genotypes and allele frequencies of 206 healthy Han Chinese in Jilin were detected and analyzed. The results showed that homozygous ε3 / 3 genotype frequency and ε3 allele frequency was the highest. ε2 is higher than ε4. Ε2 increases with age. Conclusion: The method is fast, accurate and high resolution. ApoE gene polymorphism distribution of ApoE physiological function of the study and further lay the foundation for clinical application.
