目的:探讨马蔺子素对SD大鼠放射性肺损伤的防护作用。方法:将210只SD大鼠随机分为0.9%氯化钠溶液组(正常组),马蔺子素组(单药组),单纯照射组(模型组),激素组,马蔺子素大、中、小剂量(60,30,15 mg·kg~(-1)·d~(-1))组,每组30只。采用~(60)Coγ单次20 Gy右全肺照射,建立大鼠放射性肺损伤模型。照射后第1,4,8,12,24周取大鼠肺组织苏木素伊红(HE)染色,马松三色(Masson)染色进行组织病理学观察;紫外分光光度计检测肺组织中羟脯氨酸含量;酶联免疫吸附测定法(ELISA)检测血清肿瘤坏死因子-α(TNF-α),白细胞介素-1β(IL~(-1)β)和转化生长因子-β_1(TGF-β_1)浓度;免疫组化法检测肺组织中TNF-α,TGF-β_1的表达。结果:与模型组比较,大、中剂量马蔺子素照射组可显著降低放射性肺损伤的程度,且有降低羟脯氨酸含量和抑制TNF-α,IL~(-1)β,TGF-β_1表达的作用(P<0.05)。单药组各项指标与正常组相当。结论:中、大剂量马蔺子素对放射性肺损伤有较好的防护作用,且对大鼠肺组织几乎无毒性。
Objective: To investigate the protective effect of irisquinone on radiation-induced lung injury in SD rats. Methods: 210 SD rats were randomly divided into 0.9% sodium chloride solution group (normal group), irisquinone group (single drug group), simple irradiation group (model group), hormone group, irisquinone large, medium and small Dose (60,30,15 mg · kg -1 · d -1) group, 30 rats in each group. Acute radiation-induced lung injury in rats was induced by a single (20 Gy) whole lung irradiation with ~ (60) Co γ. The lung tissues of rats were harvested at 1, 4, 8, 12 and 24 weeks after irradiation for HE staining and Masson staining for histopathology. UV spectrophotometer was used to detect the expression of Hormone Serum levels of tumor necrosis factor-α (TNF-α), interleukin-1β (IL-1β) and transforming growth factor-β1 (TGF-β1) were detected by enzyme linked immunosorbent assay (ELISA) ), And the expression of TNF-α and TGF-β_1 in lung tissues were detected by immunohistochemistry. Results: Compared with model group, large and medium dose irisquinone irradiation group could significantly reduce the degree of radiation-induced lung injury, reduce the content of hydroxyproline and inhibit the expression of TNF-α, IL-1β and TGF- (P <0.05). Monotherapy group indicators and the normal group. Conclusion: The medium and large doses of irisquinon have a good protective effect on radiation-induced lung injury and are almost non-toxic to rat lung tissue.
