[目的]探讨轮叶党参中分离到的齐墩果酸(OA)对紫外线照射所诱发DNA损伤的保护作用.[方法]取大鼠采用给脾细胞行紫外线照射的方法制作DNA损伤模型,分别给予10,20,40,80mg/L的OA;给DNA损伤大鼠脾细胞加入40mg/L齐墩果酸,培养30,60,90,120min,用荧光分光光度法检测DNA裂解率.[结果]给大鼠脾细胞行紫外线照射后,DNA裂解率明显增高(P<0.01),当OA终质量浓度分别为10,20,40,80mg/L时,紫外线照射之前用OA预处理2h可降低紫外线照射所致的DNA裂解率,DNA裂解率随OA加入量的增加而逐渐降低,当OA剂量超过20mg/L后DNA裂解率下降缓慢.单纯紫外线照射组和加入OA组的DNA裂解率均随着培养时间的延长而逐渐下降,当培养时间达到60min后,两组DNA裂解率间差异有统计学意义,加入OA组的DNA裂解率明显低于单纯紫外线照射组.[结论]齐墩果酸可保护紫外线照射所诱发的DNA损伤,且促进断裂DNA的修复过程.
[Objective] To investigate the protective effect of oleanolic acid (OA) isolated from Codonopsis pilosula on DNA damage induced by UV irradiation. [Methods] The model of DNA damage was established by exposing spleen cells to UV light. 10, 20, 40 and 80 mg / L of OA were added into the spleen cells of rats with DNA damage. 40 mg / L oleanolic acid was added to the spleen cells of rats with DNA damage, and the DNA cleavage rate was detected by fluorescence spectrophotometry after cultured for 30, 60, 90 and 120 min. [Results] When the final concentration of OA was 10, 20, 40 and 80 mg / L respectively, pretreatment of OA with UV for 2 hours reduced the UV light DNA cleavage rate and DNA cleavage rate decreased with the increase of OA dosage, but the DNA cleavage rate decreased slowly when the dose of OA exceeded 20 mg / L. The cleavage rate of DNA in both ultraviolet irradiation group and OA group increased with When the incubation time reached 60min, there was a significant difference in DNA cleavage rate between the two groups, and the DNA cleavage rate of OA group was significantly lower than that of UV alone group. [Conclusion] Oleanolic acid Protect DNA damage induced by ultraviolet radiation, and promote DNA repair process.
