近年来的研究发现,单核细胞趋化蛋白1(MCP-1)是一种极为强烈的单核细胞趋化因子。本研究着重观察了极低密度脂蛋白(VLDL)和氧化修饰的极低密度脂蛋白(OX—VLDL)对单核细胞和巨噬细胞MCP-1 mRNA表达的影响。在单核细胞和巨噬细胞的培养基中分别加入25μg/ml的VLDL或OX—VLDL,培养24h后提取单核细胞和巨噬细胞总RNA,用MCP-1寡核甘酸探针,进行Northern blot分析。结果显示单核细胞和巨噬细胞都能表达MCP-1 mRNA,OX-VLDL能增强单核/巨噬细胞MCP-1 mRNA的表达。这提示单核和巨噬细胞能促进单核细胞迁入内皮下间隙,而OX-VLDL能增强这种作用。
Recent studies have found that monocyte chemotactic protein 1 (MCP-1) is a very strong monocyte chemotactic factor. This study focused on the effects of very low density lipoprotein (VLDL) and oxidized low density lipoprotein (OX-VLDL) on MCP-1 mRNA expression in monocytes and macrophages. VLDL or OX-VLDL at 25 μg / ml was added to the culture medium of monocytes and macrophages, respectively. Total RNA of monocytes and macrophages was extracted after culturing for 24 hours, and Northern blotting was performed using MCP-1 oligonucleotide probe blot analysis. The results showed that both monocytes and macrophages could express MCP-1 mRNA. OX-VLDL enhanced MCP-1 mRNA expression in monocytes / macrophages. This suggests that monocytes and macrophages can promote monocyte migration into the subepithelial space, whereas OX-VLDL potentiates this effect.
