为探讨碳酸氢钠对嗜水气单胞菌毒力基因表达的影响,利用荧光定量PCR技术,以未添加碳酸氢钠的嗜水气单胞菌为对照组,添加0.024、0.048、0.072、0.096和0.120 mol/L碳酸氢钠的嗜水气单胞菌为实验组,对嗜水气单胞菌的3个主要毒力基因——溶血素基因(AHH-1)、气溶素基因(Aer A)和弹性蛋白酶基因(ahyB)进行相对表达分析。结果表明,5个实验组嗜水气单胞菌的AHH-1、AerA和ahyB表达量均显著低于对照组。且随着碳酸氢钠浓度的增加,基因表达呈逐渐下调趋势。人工感染结果表明,添加碳酸氢钠培养的嗜水气单胞菌对斑马鱼的半致死量明显比对照组高,而且随着碳酸氢钠浓度的增加,嗜水气单胞菌对斑马鱼的半致死量呈现逐渐加大的趋势,表明毒力逐渐减小。
In order to investigate the effect of sodium bicarbonate on the virulence gene expression of Aeromonas hydrophila, fluorescent quantitative PCR technique was used to compare the effects of sodium bicarbonate-added Aeromonas hydrophila control group, 0.024,0.048,0.072,0.096 And Aeromonas hydrophila (0.120 mol / L sodium bicarbonate) were used as experimental groups. The results of artificial infection showed that the semi-lethal dose of Aeromonas hydrophila cultured with sodium bicarbonate was significantly higher than that of the control group, and with the increase of sodium bicarbonate concentration, The lethal dose showed a gradually increasing trend, indicating that the virulence gradually decreased.
