以板栗花芽为试材,研究了建立其花芽中吲哚乙酸(IAA)、脱落酸(ABA)、玉米素核苷(ZR)、赤霉素(GA3)4种内源激素含量的定性、定量分析方法。结果表明:采用80%的预冷甲醇水溶液浸提板栗枝芽中内源激素IAA、ABA、GA3、ZR,提取效果较好;采用SPE-C18小柱固相萃取技术进行纯化,采用Symmetry 300TMC18(5μm,4.6mm×250mm),以甲醇:0.075%冰醋酸(90∶5,V∶V)为流动相,在流速1.0mL/min、柱温30℃,检测波长219nm的条件下,进行梯度洗脱,分离效果理想,加标回收率达到89.92%~105.30%;该方法准确、灵敏、重现性好。
