目的:探讨ACL重建及半月板修复所用异体移植物的组织安全性。方法:分别以不同浓度梯度人体骨-髌腱-骨和半月板移植物浸提液培养NIH3T3细胞,光镜下观察细胞形态,MTT法检测受试物中存活细胞百分比,其结果与DMEM培养液(阴性对照组)及苯酚(阳性对照组)中存活细胞百分比进行比较。结果:光镜下各种浓度移植物浸提液培养的细胞形态均正常,存活率均大于97%,明显优于阳性对照组。结论:本实验中人体骨-髌腱-骨和半月板移植物未见细胞毒性。
OBJECTIVE: To investigate the tissue safety of allografts used for ACL reconstruction and meniscal repair. Methods: NIH3T3 cells were cultured in bone marrow-patellar tendon-bone and meniscus implants with different concentration gradient respectively. The cell morphology was observed under light microscope. The percentage of viable cells in the test substance was detected by MTT assay. The results were compared with that of DMEM medium (Negative control group) and phenol (positive control group) were compared. Results: The morphology of the cells cultured in the extracts of various concentrations under the light microscope was normal, with the survival rates of more than 97%, which were significantly better than the positive control group. CONCLUSION: No cytotoxicity has been observed in human bone-patellar tendon-bone and meniscus implants in this experiment.
