应用调节性T细胞(regulatory T cell,Treg)治疗自身免疫性疾病、移植排斥反应和移植物抗宿主病等疾病的细胞疗法目前正在进行临床试验。Treg细胞的稳定性是应用Treg细胞进行细胞治疗的一个非常关键的问题。叉头转录因子Foxp3在Treg细胞发育和维持过程中起重要的作用,Foxp3的突变可引起Treg细胞的功能障碍。本综述对影响Treg细胞稳定性的各种因素进行探讨,包括Foxp3的转录水平的调控、表观遗传学调控及翻译后修饰的调控,为获得稳定的Treg细胞并更好地应用于相关免疫疾病的治疗提供新思路。“,”Cell therapies using regulatory T cells (Treg) to treat diseases such as autoimmune diseases, transplant rejection, and graft-versus-host disease are currently undergoing clinical trials. The stability of Treg cells is a critical issue for Treg cells for cell therapy. The forkhead transcription factor 3 (Foxp3) plays an important role in the development and maintenance of Treg cells. Mutations of Foxp3 can cause dysfunction of Treg cells. This review discusses various factors that affect the stability of Treg cells, including the regulation of the transcription level, the regulation of epigenetics, and the regulation of post-translational modifications of Foxp3, in order to provide new ideas for using stable Treg cells in the treatment of such immune diseases.
