目的了解广州市男男性行为者(MSM)Ⅰ型艾滋病病毒(HIV-1)亚型的流行状况。方法将2011年广州市发现的227份经男男性行为感染的HIV-1阳性血清样本,应用巢式-聚合酶链反应(Nested-PCR)扩增gag区和env区基因,测序和分析亚型。结果成功扩增了213份样本的env区基因序列和214份样本的gag区基因序列。经过系统进化树分析并综合考虑两个区的亚型结果,发现成功扩增的220份样本中,CRF01_AE重组型98份(44.5%),CRF07_BC重组型87份(39.5%),CRF59_01B重组型3份(1.4%),B亚型16份(7.3%),C亚型3份(1.4%),A亚型1份(0.5%),有12份样本无法确定亚型。env区各亚型组内基因离散率:CRF01_AE重组型(12.8%±0.9%),CRF07_BC(11.5%±1.3%),CRF59_01B(7.4%±1.0%),B亚型(19.6%±1.3%)。gag区各亚型基因离散率:CRF01_AE重组型(6.2%±0.5%),CRF07_BC(3.4%±0.3%),CRF59_01B(2.1%±0.5%),B亚型(8.4%±0.8%)。结论广州市MSM HIV-1感染人群中存在着CRF01_AE、CRF07_BC、CRF59_01B三种重组型和B、C、A三种亚型,CRF01_AE、CRF07_BC重组型为优势流行株。CRF59_01B重组型可能是MSM近年来新出现的流行株。
Objective To understand the prevalence of HIV-1 subtype of MSM in Guangzhou. Methods HIV-1 positive sera from 227 male patients were detected by Nested-PCR in 2011 in Guangzhou. The gag region and env region were amplified by nested-PCR, and the subtype was sequenced and analyzed . Results The env region of 213 samples and the gag region of 214 samples were successfully amplified. After analyzing phylogenetic tree and considering the subtype results of the two regions, we found that 98 (44.5%) CRF01_AE, 87 (39.5%) CRF07_BC, CRF59_01B (1.4%), 16 subtype B (7.3%), 3 subtype C (1.4%) and 1 subtype A (0.5%). In the env region, the gene dispersal rate of CRF01_AE (12.8% ± 0.9%), CRF07_BC (11.5% ± 1.3%), CRF59_01B (7.4% ± 1.0%) and B subtype (19.6% ± 1.3% . The genetic divergence rate of each subtype of gag region was 6.2% ± 0.5% of CRF01_AE, 3.4% ± 0.3% of CRF07_BC, 2.1% ± 0.5% of CRF59_01B and 8.4% ± 0.8% of B subtype. Conclusion There are three recombinant CRF01_AE, CRF07_BC and CRF59_01B and three subtypes B, C and A in MSM HIV-1 infected population in Guangzhou. CRF01_AE and CRF07_BC recombinants are predominant epidemic strains. CRF59_01B recombinant may be a new epidemic strain of MSM in recent years.
