目的:评价Period2(Per2)基因转染后过表达对人宫颈癌细胞(Hela)X射线放射敏感性的影响。方法:采用脂质体介导方法将Per2基因转染至Hela细胞,以空白组及空载体质粒pcDNA3.1(+)为对照,采用Wsetern Blot检测Per2基因在Hela细胞中的表达,给予X射线单次照射后,流式细胞仪检测细胞凋亡,采用细胞集落形成实验检测细胞增殖情况。结果:X射线照射后,与转染空载体质粒组及空白组比较,转染Per2基因Hela细胞凋亡率增加,细胞克隆形成减少。结论:节律基因Per2过表达使X射线照射的人宫颈癌Hela细胞凋亡增加,细胞增殖减弱,明显增强了HeLa细胞对放射损伤的敏感性。
Objective: To evaluate the effect of Per2 gene transfection on the radiosensitivity of human cervical carcinoma cells (Hela) after transfection. Methods: The Per2 gene was transfected into Hela cells by liposome-mediated method. The expression of Per2 gene in Hela cells was detected by Wsetern Blot using pcDNA3.1 (+) blank vector and empty plasmid as control. X-ray After a single irradiation, apoptosis was detected by flow cytometry, and cell proliferation was detected by colony formation assay. Results: After X-ray irradiation, the apoptosis rate of Hela cells transfected with Per2 gene was increased and the formation of cell clone was decreased compared with the blank plasmid transfected group and the blank group. Conclusion: Overexpression of circadian gene Per2 enhances the apoptosis of Hela cells irradiated by X-rays and attenuates the proliferation of Hela cells. The sensitivity of HeLa cells to radiation injury is significantly enhanced.
