目的观察口服低剂量混合稀土“常乐”对糖尿病大鼠心肌NF-KB、TNF-α表达的影响。方法以Wistar雄性大鼠45只为研究对象,取35只造模,造模成功后,将实验动物分为3组即对照组(10只)、糖尿病模型组(10只)和混合稀土“常乐”给药组(10只),3组大鼠分别每日给予生理盐水、生理盐水、混合稀土“常乐”(0.2 mg/kg)灌胃1个月,然后处死取血液和心脏,采用放免法检测大鼠血浆中血糖、甘油三酯(TG)、总胆固醇(TC)、NF-KB及TNF-α的含量;RTPCR法检测大鼠心肌组织中NF-KB及TNF-α的mRNA表达水平;免疫组化法检测大鼠心肌组织中NF-KB及TNF-α的蛋白表达;苏木素-伊红(HE)染色光学显微镜下观察心肌结构。结果糖尿病模型组大鼠血浆中血糖、甘油三酯(TG)、总胆固醇(TC)、NF-KB及TNF-α的含量明显高于对照组和混合稀土“常乐”给药组且大鼠心肌组织中NF-KB及TNF-α的mRNA水平和NF-KB及TNF-α的蛋白水平也明显高于对照组和混合稀土“常乐”给药组;光学显微镜下可见对照组大鼠心肌细胞排列紧密整齐、糖尿病组大鼠心肌细胞排列疏散紊乱间质可见炎细胞浸润、混合稀土“常乐”组大鼠心肌细胞基本整齐紧密。结论口服混合稀土“常乐”(0.2 mg/kg)可抑制糖尿病大鼠NF-KB、TNF-α的表达对糖尿病大鼠心肌组织有一定的保护作用。

The myocardial cells in diabetic rats were arranged disorganized interstitial infiltration of inflammatory cells. The myocardial cells in mixed rare earth “Changle ” group were basically neat and tidy. Conclusion Oral mixed rare earth “Changle” (0.2 mg / kg) can inhibit the expression of NF-κB and TNF-α in diabetic rats to some extent.
