口服低剂量混合稀土“常乐”对糖尿病大鼠心肌NF-KB和TNF-α表达的影响

目的观察口服低剂量混合稀土“常乐”对糖尿病大鼠心肌NF-KB、TNF-α表达的影响。方法以Wistar雄性大鼠45只为研究对象,取35只造模,造模成功后,将实验动物分为3组即对照组(10只)、糖尿病模型组(10只)和混合稀土“常乐”给药组(10只),3组大鼠分别每日给予生理盐水、生理盐水、混合稀土“常乐”(0.2 mg/kg)灌胃1个月,然后处死取血液和心脏,采用放免法检测大鼠血浆中血糖、甘油三酯(TG)、总胆固醇(TC)、NF-KB及TNF-α的含量;RTPCR法检测大鼠心肌组织中NF-KB及TNF-α的mRNA表达水平;免疫组化法检测大鼠心肌组织中NF-KB及TNF-α的蛋白表达;苏木素-伊红(HE)染色光学显微镜下观察心肌结构。结果糖尿病模型组大鼠血浆中血糖、甘油三酯(TG)、总胆固醇(TC)、NF-KB及TNF-α的含量明显高于对照组和混合稀土“常乐”给药组且大鼠心肌组织中NF-KB及TNF-α的mRNA水平和NF-KB及TNF-α的蛋白水平也明显高于对照组和混合稀土“常乐”给药组;光学显微镜下可见对照组大鼠心肌细胞排列紧密整齐、糖尿病组大鼠心肌细胞排列疏散紊乱间质可见炎细胞浸润、混合稀土“常乐”组大鼠心肌细胞基本整齐紧密。结论口服混合稀土“常乐”(0.2 mg/kg)可抑制糖尿病大鼠NF-KB、TNF-α的表达对糖尿病大鼠心肌组织有一定的保护作用。 Objective To observe the effect of low dose mixed rare earth “Changle ” on the expression of NF-KB and TNF-|Áin myocardium of diabetic rats. Results The levels of plasma glucose, triglyceride (TG), total cholesterol (TC), NF-κB and TNF-α in diabetic model group were significantly higher than those in control group and mixed rare earth The mRNA level of NF-κB and TNF-α and the protein level of NF-κB and TNF-α in myocardial tissue of rats were also significantly higher than that of the control group and the mixed rare earth “Changle” group. Under the light microscope, The myocardial cells of rats in group A were arranged neatly and neatly. The myocardial cells in diabetic rats were arranged disorganized interstitial infiltration of inflammatory cells. The myocardial cells in mixed rare earth “Changle ” group were basically neat and tidy. Conclusion Oral mixed rare earth “Changle” (0.2 mg / kg) can inhibit the expression of NF-κB and TNF-α in diabetic rats to some extent.
