目的:探讨青蒿素对狼疮性肾炎小鼠外周血单个核细胞GRαmRNA、GRβmRNA及肾组织P300/CBP蛋白表达的影响。方法:40只雌性B6D2F1代杂交小鼠随机分为正常对照组、模型组、泼尼松组(泼尼松混悬液灌养:6.45 mg/kg)及青蒿素组(青蒿素混悬液灌养:150 mg/kg)。通过诱导方法建立狼疮性肾炎小鼠模型,分别采用RT-PCR及免疫组化(SP)法等检测技术测定外周血单个核细胞GRα mRNA、GRβ mRNA表达及肾组织转录共激活因子P300/CBP蛋白表达情况,观察青蒿素对上述指标的影响。结果:与模型组比较,青蒿素组动物外周血单个核细胞GRαmRNA和肾组织转录共激活因子P300/CBP蛋白表达均有不同程度升高(P<0.01),GRβ mRNA表达显著降低(P<0.05或P<0.01),且青蒿素对GR αmRNA和转录共激活因子P300/CBP蛋白表达的影响明显优于泼尼松组(P<0.01)。结论:青蒿素治疗狼疮性肾炎的作用机制可能与调节外周血单个核细胞GRα mRNA、GRβ mR-NA及肾组织P300/CBP蛋白表达有关。
Objective: To investigate the effect of artemisinin on GRαmRNA, GRβmRNA and P300 / CBP protein expression in peripheral blood mononuclear cells of lupus nephritis mice. Methods: Forty female B6D2F1 hybrid mice were randomly divided into normal control group, model group, prednisone group (prednisone suspension: 6.45 mg / kg) and artemisinin group (artemisinin suspension Liquid feeding: 150 mg / kg). The mouse model of lupus nephritis was established by induction method. The expression of GRα mRNA, GRβ mRNA and the expression of P300 / CBP protein in peripheral blood mononuclear cells were determined by RT-PCR and immunohistochemistry (SP) Expression, observe the impact of artemisinin on the above indicators. Results: Compared with the model group, the expression of GRαmRNA and P300 / CBP protein in artemisinin group increased to different extents (P <0.01) and GRβ mRNA expression decreased significantly (P < 0.05 or P <0.01), and the effect of artemisinin on GRαmRNA and P300 / CBP protein expression was significantly better than prednisone group (P <0.01). Conclusion: The mechanism of artemisinin treatment of lupus nephritis may be related to the regulation of GRα mRNA, GRβ mR-NA and P300 / CBP protein expression in peripheral blood mononuclear cells.
