目的 :探讨大鼠生后下丘脑不同发育阶段细胞凋亡的规律及禁水对细胞凋亡的影响。方法 :取生后发育不同阶段的大鼠下丘脑 ,①用Bouin液固定 ,用TUNEL染色 ;②用改良的Bouin液固定 ,核固红 结晶紫染色。结果 :①生后 1d凋亡细胞最多 ,主要位于下丘脑背侧室旁核附近。生后 5d ,凋亡细胞明显减少。以后凋亡细胞越来越少。 60d极少见到凋亡细胞。两种染色方法结果相似 ;②成年大鼠禁水后 ,未见凋亡细胞增多。结论 :①细胞凋亡受基因和细胞外环境因素的共同调节 ;②禁水对大鼠下丘脑细胞凋亡无影响
OBJECTIVE: To investigate the regularity of apoptosis in hypothalamus at different developmental stages and the effect of water forbidding apoptosis. METHODS: The hypothalamus of rats in different stages of development was taken after birth. ① The cells were fixed with Bouin solution and stained with TUNEL. ② The cells were fixed with modified Bouin solution and stained with red crystal violet. Results: ① The apoptotic cells were most abundant at 1 day after birth, mainly located in the dorsal paraventricular nucleus of the hypothalamus. 5d after birth, apoptotic cells decreased significantly. After the apoptotic cells less and less. 60d rarely seen apoptotic cells. The results of the two staining methods were similar; ②When the water of the adult rats were not in water, no apoptotic cells were found. Conclusion: ①Apoptosis is regulated by gene and extracellular environment factors; ②Forbidden water has no effect on apoptosis of hypothalamus in rats
