目的:采用遗传算法优化猪苓多糖的提取工艺,进一步研究其体外抗氧化活性。方法:选择溶媒倍数、提取温度、提取时间和提取次数为考察因素,在正交试验基础上采用遗传算法优化猪苓多糖的提取工艺;根据Fenton反应和DPPH法进行抗氧化活性测定。结果:最佳提取工艺为:加药材41量的水,于99℃提取3次,每次2.5 h,猪苓多糖提取率为17.73 mg/g;猪苓多糖对DPPH及·OH自由基有较高的清除率,且清除率与浓度呈量效关系。结论:遗传算法优化所得提取工艺具有提取率高、稳定的优点,可用于猪苓多糖的提取;猪苓多糖具有良好的体外抗氧化活性。
OBJECTIVE: To optimize the extraction process of Polyporus umbellatus polysaccharide by genetic algorithm and further study its antioxidant activity in vitro. Methods: The solvent multiples, extraction temperature, extraction time and number of extraction were selected as the investigation factors. Based on the orthogonal test, genetic algorithm was used to optimize the extraction process of Polyporus umbellatus polysaccharide. The antioxidant activity was determined by Fenton reaction and DPPH method. Results: The optimum extraction process was as follows: water of medicinal material 41 was extracted three times at 99 ℃ for 2.5 h, the extraction rate of Polyporus umbellatus polysaccharide was 17.73 mg / g; Polyporus umbellatus polysaccharide had more effect on DPPH and · OH free radical High clearance rate, and the clearance rate and the concentration was dose-effect relationship. Conclusion: The genetic algorithm optimized extraction process has the advantages of high extraction rate, stability, can be used for the extraction of Polyporus umbellatus; Polyporus umbellatus polysaccharide has good in vitro antioxidant activity.
