目的检测微小核糖核酸(microRNA,miR)-200b在肾癌患者血清中的表达状况,同时研究miR-200b与肾癌患者临床病理特征之间的关联,为进一步的深入研究奠定基础。方法以2014年1月至2015年12月初次就诊并诊断为肾细胞癌的患者(术后病理证实为肾癌)35例为肾癌组,35例健康体检者(各项指标均正常)为健康对照组。收集肾癌组患者术前血清样本及健康对照组对象的血清样本,利用实时荧光定量逆转录-聚合酶链反应(RTq PCR)检测和对比miR-200b在两组对象血清中的相对表达量,分析肾癌患者临床病理特征和miR-200b相对表达量之间的关系。结果肾癌患者miR-200b相对表达量为3.24±1.72,健康对照患者相对表达量为7.39±3.58,两组差异有统计学意义(P<0.05)。与健康对照组比较,35例肾癌中有29例(82.86%)miR-200b表达下调。肾癌患者的性别、年龄、肾癌组织类型、肾肿瘤大小、TNM分期和其血清中miR-200b相对表达量无相关性(P均>0.05)。结论 miR-200b在肾癌患者血清中低表达,提示其可能与肾癌的发生发展相关。
Objective To detect the expression of microRNA (miR) -200b in the serum of patients with renal cell carcinoma, and to study the relationship between miR-200b and clinicopathological features of patients with renal cell carcinoma, which will lay the foundation for further study. Methods From January 2014 to December 2015, 35 patients with primary renal cell carcinoma who were diagnosed as renal cell carcinoma (pathologically confirmed as renal cell carcinoma) and 35 healthy controls (all indicators were normal) were Healthy control group. Serum samples from patients with renal cell carcinoma before operation and healthy controls were collected and the relative expression levels of miR-200b in serum of two groups were detected by real-time fluorescent quantitative reverse transcription-polymerase chain reaction (RT-PCR) The relationship between the clinicopathological features and the relative expression of miR-200b in renal cell carcinoma was analyzed. Results The relative expression level of miR-200b was 3.24 ± 1.72 in renal cell carcinoma patients and 7.39 ± 3.58 in healthy controls. The difference between the two groups was statistically significant (P <0.05). Compared with healthy controls, miR-200b was down-regulated in 29 of 35 (82.86%) renal cancers. There was no correlation between gender, age, type of renal cell carcinoma, size of renal tumor, TNM stage and the relative expression of miR-200b in serum (all P> 0.05). Conclusion The low expression of miR-200b in the serum of patients with renal cell carcinoma suggests that it may be related to the occurrence and development of renal cell carcinoma.
