目的:探讨人参皂苷Rg3对人膀胱癌细胞增殖作用的影响及作用机制。方法:采用人膀胱癌T24细胞株进行细胞培养,将人参皂苷Rg3分别以0,5,10,20和40μmol·L~(1-)的剂量处理细胞24 h后,采用四甲基偶氮唑盐(MTT)方法检测人参皂苷Rg3对人膀胱癌T24细胞增殖的抑制作用,倒置显微镜和流式细胞术观察人参皂苷Rg3对T24细胞凋亡的诱导作用,应用RT-PCR和Western blot方法检测不同浓度人参皂苷Rg3处理后T24细胞中EphB4 mRNA及其蛋白的表达情况。结果:人参皂苷Rg3对膀胱癌T24细胞具有较强的抑制作用,且这种抑制作用随浓度和时间增加而增大,呈浓度依赖关系(P<0.05)。不同浓度人参皂苷Rg3作用下的膀胱癌T24细胞中,EphB4 mRNA及其蛋白的表达明显减弱,且呈剂量梯度下降(P<0.05)。结论:人参皂苷Rg3的抗癌活性与其抑制EphB4表达有关。
Objective: To investigate the effect of ginsenoside Rg3 on human bladder cancer cell proliferation and its mechanism. Methods: Human bladder cancer cell line T24 was used for cell culture. Ginsenoside Rg3 was treated with 0, 5, 10, 20 and 40 μmol·L ~ (-1) for 24 h, (MTT) assay was used to detect the inhibitory effect of ginsenoside Rg3 on the proliferation of human bladder cancer T24 cells. The apoptosis of T24 cells induced by ginsenoside Rg3 was observed by inverted microscopy and flow cytometry. The expression of ginsenoside Rg3 was detected by RT-PCR and Western blot The expression of EphB4 mRNA and its protein in T24 cells treated with ginsenoside Rg3. Results: Ginsenoside Rg3 had a strong inhibitory effect on bladder cancer T24 cells. The inhibitory effect of Ginsenoside Rg3 increased with concentration and time increasing in a concentration-dependent manner (P <0.05). The expression of EphB4 mRNA and its protein in bladder cancer T24 cells treated with different concentrations of ginsenoside Rg3 decreased significantly, and the dose gradient decreased (P <0.05). Conclusion: The anti-cancer activity of ginsenoside Rg3 is related to the inhibition of EphB4 expression.
