目的 建立高效液相色谱法测定大鼠血浆中丹参酮IIA浓度的方法。方法 血浆样品经液 液萃取后 ,用HPLC法进行分析。色谱柱为YMCC1 8(5 μm ,ID 3 0mm× 1 50mm) ;流动相为乙腈 水 冰醋酸 (74∶2 6∶1 ) ;流速 0 3mL·min-1 ;检测波长 2 70nm ;内标为 4 氯联苯。结果 线性范围为 0 0 5mg·L-1 ~ 6 40mg·L-1 ,最低检测浓度为 0 0 5mg·L-1 。高、中、低 3种浓度的平均方法回收率分别为 98 9% ,1 0 2 1 %和 1 0 0 4%。日内、日间精密度 (RSD)均小于5 %。结论 本方法稳定、简便、可靠 ,可用于丹参酮IIA的血药浓度分析及其药代动力学研究
Objective To establish a method for the determination of tanshinone IIA in rat plasma by high performance liquid chromatography. Methods Plasma samples were extracted with liquid and analyzed by HPLC. The column was YMCC18 (5 μm, ID30mm×150mm); the mobile phase was acetonitrile glacial acetic acid (74∶2 6∶1); the flow rate was 0.3mL·min-1; the detection wavelength was 2 70nm; the internal standard was 4 Chlorobiphenyl. The linear range was 0 0 5mg·L -1 to 6 40mg·L -1 , and the lowest concentration was 0 0 5mg·L -1 . The average method recoveries for high, medium, and low concentrations were 98 9%, 10 21%, and 100%, respectively. The intraday and inter-day precision (RSD) were less than 5%. Conclusion This method is stable, simple and reliable. It can be used for the analysis of blood concentration of tanshinone IIA and its pharmacokinetics.
