采用响应面法优化冬虫夏草CS.SYSU-Ⅱ胞内多糖提取工艺。在单因素试验基础上,固定提取次数,选取液料比、提取时间、提取温度为自变量,多糖提取率为响应值,利用Box-Behnken中心组合设计原理和响应面分析法确定最佳提取工艺条件。结果表明,热水浸提法提取胞内多糖的最佳工艺参数为:提取温度96.99℃、提取时间4.38 h、液料比35.78︰1(mL/g)、浸提次数2次,在此条件下,多糖提取率为5.47%。与理论预测值相比,其相对误差为0.15%。运用响应面分析法得出的冬虫夏草CS.SYSU-Ⅱ胞内多糖的提取工艺具有一定的实际应用价值。
Optimization of Extraction Process of Intracellular Polysaccharide of Cordyceps Sinensis CS.SYSU-Ⅱ by Response Surface Methodology. Based on the single-factor experiments, the optimal extraction conditions were determined by the method of Box-Behnken central composite design and response surface analysis, with the number of fixed extraction, liquid-solid ratio, extraction time, extraction temperature as independent variables and polysaccharide extraction rate as response values condition. The results showed that the optimum technological parameters for extraction of intracellular polysaccharides by hot water extraction were extraction temperature 96.99 ℃, extraction time 4.38 h, liquid to liquid ratio 35.78︰1 (mL / g) and extraction times 2 times. Under these conditions Under the polysaccharide extraction rate of 5.47%. Compared with the theoretical prediction, the relative error is 0.15%. The response surface analysis of Cordyceps Sinensis CS.SYSU-Ⅱ intracellular polysaccharide extraction process has some practical value.
