目的优化一种甲型H1N1流感的双重荧光PCR快速筛查方法。方法根据WHO推荐的甲型H1N1流感检测方案合成、标记引物和探针。通过改进反应条件与参数,优化了甲型H1N1流感的双重荧光PCR检测方法。结果该方法敏感度高,特异性强,重复性好。应用该方法检测患者实际样品,结果与WHO推荐方法的检测结果一致;应用该法可在4h内完成对样品中甲型H1N1流感的检验。结论本研究优化的甲型H1N1流感的双重荧光PCR快速筛查方法可用于甲型H1N1流感的诊断。
Objective To optimize a rapid double-fluorescent PCR assay for influenza A (H1N1). Methods According to the WHO recommended influenza A H1N1 influenza detection program synthesis, labeling primers and probes. By improving the reaction conditions and parameters, we optimized the double fluorescent PCR detection method of influenza A (H1N1). Results The method is sensitive, specific and reproducible. The method was applied to detect the actual samples of patients and the results were consistent with the results of the WHO recommended method. The application of this method can complete the test of influenza A (H1N1) in samples within 4 hours. Conclusion The double fluorescent PCR rapid screening method optimized for this study can be used for the diagnosis of influenza A (H1N1).
