目的比较不同品系及相同品系不同来源BALB/c小鼠对戊型肝炎疫苗(HE疫苗)的免疫应答,探讨适用于HE疫苗评价的小鼠品系,规范疫苗效力评价用动物。方法分别用2家企业制备的HE疫苗免疫BALB/c、C57BL/6、KM和NIH 4种品系小鼠及3种不同公司来源的BALB/c小鼠,均免疫1针,28 d后采血,分离血清,检测抗体应答,计算效力ED_(50)。结果 2家疫苗免疫C57BL/6、KM和NIH小鼠时,各剂量组均表现出敏感应答,提示C57BL/6、KM和NIH小鼠对本研究中的2家疫苗不能灵敏地反映出剂量变化趋势,不适用于HE疫苗效力评价;BALB/c小鼠对2家企业各剂量组HE疫苗均呈现良好的免疫应答,且呈明显的剂量效应关系。3种不同来源的BALB/c小鼠免疫HE疫苗后,抗体应答存在差异。结论 BALB/c小鼠可作为评价HE疫苗免疫原性和效力ED_(50)的模型动物;应用BALB/c小鼠进行疫苗评价时需固定小鼠来源。
Objective To compare the immune responses of different strains and different strains of BALB / c mice to hepatitis E vaccine (HE vaccine) in different strains, and to explore the mouse strains suitable for HE vaccine evaluation and to standardize the vaccine efficacy evaluation animal. Methods BALB / c, C57BL / 6, KM and NIH strains of BALB / c mice and BALB / c mice of 3 different companies were immunized with 2 HE vaccines. Serum was separated and antibody response was measured to calculate the potency ED_ (50). Results When C57BL / 6, KM and NIH mice were immunized with the two vaccines, all the doses showed sensitive responses, suggesting that the two vaccines in C57BL / 6, KM and NIH mice could not sensitively reflect the dose trend , Not suitable for the evaluation of HE vaccine efficacy; BALB / c mice showed good immune response to HE vaccines of two companies with dose-effect relationship. BALB / c mice from 3 different sources of HE vaccination vaccine, antibody response differences. Conclusions BALB / c mice can be used as model animals to evaluate the immunogenicity and potency of HE vaccine ED_ (50). Mouse BALB / c mice need to be sourced for vaccine evaluation.
