目的 探讨国产降纤酶对大鼠局灶性脑缺血 /再灌注动物模型的神经行为学和脑梗死体积的影响 ,以证明其是否有脑保护作用。方法 采用线栓法制作大鼠大脑中动脉闭塞模型 ,用 Zea L onga 5分制评分和 TTC染色法评价神经行为学和脑梗死体积。由吉林大学第一医院和复旦大学华山医院共同进行实验研究 ,每部分实验动物各做一半。动物随机分为缺血 3h及再灌注 3h、6 h、2 4h和 72 h;缺血 6 h及再灌注 3h、6 h、2 4h;缺血 2 4h等共 10组。降纤酶采用 8U / kg腹腔注射给药。结果 行为学结果 :在 10组中仅于缺血 6 h再灌注 6 h组 Zea L onga评分有明显改善 (P<0 .0 5 ) ,其余各组与盐水对照组比较无显著性差异。梗死体积测定结果 :在缺血 3h各组中 ,只有缺血3h再灌注 72 h这个时间点比盐水组无明显缩小 (P>0 .0 5 ) ,其余各时间点梗死体积均明显缩小 (P分别 <0 .0 5、0 .0 1和 0 .0 0 1)。在缺血 6 h和 2 4h组中 ,各时间点与盐水组比较均明显缩小 (分别为 P<0 .0 1和 P<0 .0 5 )。结论 国产降纤酶对大鼠局灶性脑缺血及再灌注损伤有一定的保护作用。
Objective To investigate the effect of domestic defibrase on the neurobehavioral and cerebral infarction volume of focal cerebral ischemia / reperfusion model in rats to prove whether it has cerebral protective effect. Methods The rat model of middle cerebral artery occlusion was established by thread occlusion. Neurobehavioral and cerebral infarction volumes were assessed by Zea L onga 5-point score and TTC staining. First Hospital of Jilin University and Huashan Hospital of Fudan University jointly conducted an experimental study, each part of the experimental animals to do half. The animals were randomly divided into 10 groups: ischemia 3h and reperfusion 3h, 6h, 24h and 72h; ischemia 6h and reperfusion 3h, 6h, 24h; ischemia 24h. Defibrase by 8U / kg intraperitoneal injection. Results Behavioral results: In the 10 groups, the Zea Longa score was significantly improved only 6 h after ischemia and 6 h after reperfusion (P <0.05). There was no significant difference between the other groups and the saline control group. The result of infarction volume measurement showed that in the 3h ischemia group, infarction volume was significantly reduced at 3h after reperfusion for 72h (P> 0.05), while P Respectively <0.05,0 .0 1 and 0 .0 0 1). In the 6 h and 24 h ischemia groups, all the time points were significantly reduced compared with the saline group (P <0.01 and P <0.05, respectively). Conclusion Domestic defibrase has a protective effect on focal cerebral ischemia and reperfusion injury in rats.
