目的:观察扶正抑癌方含药血清对结肠癌lovo细胞侵袭转移能力及TGF-β_1和TβRⅡ蛋白表达的影响,以探讨扶正抑癌方降低结肠癌转移的机制。方法:lovo细胞体外培养,制备大鼠不同浓度含药血清后,MTT测定不同浓度含药血清对细胞增殖的影响,以5-Fu作为阳性对照药;通过细胞迁移实验检测细胞运动能力,肿瘤细胞体外侵袭实验检测细胞侵袭能力,SABC免疫组化法检测TGF-β_1和TβRⅡ的蛋白表达。结果:高浓度含药血清24 h对lovo细胞作用有效,与对照组和空白组相比,扶正抑癌方含药血清能显著降低细胞的运动能力和侵袭能力,免疫组化提示扶正抑癌方含药血清能降低lovo细胞TGF-β_1的蛋白表达和提高TβRⅡ的蛋白表达,差异具有统计学意义(P<0.01)。结论:扶正抑癌方含药血清能有效抑制lovo细胞的增值和降低其侵袭力和运动能力,初步认为其作用机制与降低lovo细胞TGF-β_1的表达和升高βRⅡ的蛋白表达有一定的相关性。
Objective: To observe the influence of Fuzheng Suppurative Decoction-containing serum on invasion and metastasis of colon lovo cells and the expression of TGF-β1 and TβRⅡ protein in order to explore the mechanism of Fuzheng Suppressor Decoction in reducing colon cancer metastasis. Methods: The lovo cells were cultured in vitro. After the rat serum was prepared at different concentrations, MTT assay was used to determine the effect of serum containing different concentrations on cell proliferation. 5-Fu was used as a positive control. Cell migration was measured by cell migration assay. In vitro invasion assay was used to detect the cell invasion ability. The protein expression of TGF-β 1 and TβRⅡ was detected by SABC immunohistochemistry. Results: Compared with the control group and the blank group, the drug-containing serum of Fuzheng Suppressing Decoction could significantly reduce the cell motility and invasion ability of the LoVo cells at 24 h. The results of immunohistochemistry showed that Fuzheng Tiao Huo Fang Drug-containing serum could reduce the protein expression of TGF-β_1 and increase the protein expression of TβRⅡ in lovo cells, with statistical significance (P <0.01). CONCLUSION: Fuzheng Suppressant Decoction-containing serum can effectively inhibit the proliferation of LoVo cells and reduce its invasiveness and motor ability, and its mechanism of action is believed to be related to the decrease of the expression of TGF-β 1 and the increase of βRⅡ protein in lovo cells Sex.
