为探讨细胞视黄醇结合蛋白(CRBP)在胃癌及癌前病变中的表达及其与维生素A水平的关系,采用SP免疫组化法和双抗体夹心ELISA法观察CRBP表达。结果CRBP阳性率在肠腺化生、不典型增生、胃癌分别为40.6%、43.3%、67.2%,均高于对照组8.3%(P<0.05、0.05、0.01),胃癌高于癌前病变(P<0.05),高、中分化胃癌高于低、未分化者(P<0.01)。胃癌组织CRBP水平高于非癌胃粘膜(P<0.01),维生素A水平低于非癌胃粘膜(P<0.01)。提示在胃癌前病变阶段就开始有维生素A代谢改变,胃癌组织CRBP增高与肿瘤细胞内维生素A水平降低有关,检测CRBP表达对选用维甲类药物治疗可能有指导意义。
To investigate the expression of retinol binding protein (CRBP) in gastric cancer and precancerous lesions and its relationship with vitamin A level, SP immunohistochemistry and double antibody sandwich ELISA were used to observe the expression of CRBP. Results The positive rates of CRBP were 40.6%, 43.3%, and 67.2% in intestinal metaplasia, atypical hyperplasia, and gastric cancer, respectively, which were significantly higher than those in the control group (8.3%, P<0.05, 0.05, and 0.01). Gastric cancer was higher than precancerous lesions ( P <0.05), high and moderately differentiated gastric cancer were higher than those of low and undifferentiated (P <0.01). The level of CRBP in gastric cancer was higher than that in non-cancerous gastric mucosa (P<0.01), and the level of vitamin A was lower than that in non-cancerous gastric mucosa (P<0.01). It is suggested that vitamin A metabolism changes at the stage of gastric precancerous lesion. The increase of CRBP in gastric cancer tissue is related to the decrease of vitamin A level in tumor cells. The detection of CRBP expression may have guiding significance for the selection of retinoid drugs.
