目的研究NIMA相关蛋白激酶6(NEK6)在肝癌(HCC)组织中的表达及意义。方法采用荧光定量PCR检测168例HCC及癌旁组织中NEK6mRNA表达,分析其与HCC临床病理参数的相关性。通过转染NEK6过表达质粒构建稳定表达NEK6的HCC细胞株,行裸鼠皮下种植,观察NEK6对HCC细胞致瘤能力的影响。结果与癌旁组织比较,HCC癌组织NEK6mRNA阳性表达率高[23.8%(40/168)vs.76.2%(128/168)](P<0.01);NEK6mRNA表达量与肿瘤大小、脉管侵犯及病理分级显著相关(P<0.05)。NEK6过表达可明显促进肝癌细胞株Huh7和HepG2的成瘤能力。结论 HCC组织高表达NEK6,且与HCC恶性程度明显相关,可促进HCC细胞成瘤。
Objective To study the expression and significance of NIMA-associated protein kinase 6 (NEK6) in hepatocellular carcinoma (HCC). Methods The expression of NEK6 mRNA in 168 HCC tissues and adjacent normal tissues was detected by real-time PCR. The correlation between NEK6 mRNA and clinical pathological parameters was analyzed. HCC cells stably expressing NEK6 were constructed by transfection of NEK6 overexpression plasmid and subcutaneously implanted into nude mice to observe the effect of NEK6 on tumorigenicity of HCC cells. Results The positive rate of NEK6 mRNA in HCC tissues was significantly higher than that in adjacent non-cancerous tissues [23.8% (40/168) vs 76.2% (128/168)] (P <0.01). The expression of NEK6 mRNA correlated with tumor size, Pathological grade was significantly correlated (P <0.05). NEK6 overexpression significantly promoted tumorigenicity of Huh7 and HepG2 cells. Conclusion High expression of NEK6 in HCC tissues is closely related to the degree of malignancy of HCC, which can promote the tumorigenesis of HCC.
