利用微生物转化的方法,选取灰色链霉菌(Streptomyces griseus ATCC 13273)为转化菌株,对去氢骆驼蓬碱的转化进行研究。该菌株可以将去氢骆驼蓬碱转化为两种转化产物(记为H1和H2),通过MS、1H-NMR和13C-NMR分析,转化产物分别鉴定为哈尔酚和N-羟基去氢骆驼蓬碱。并对转化条件进行初步优化,优化后的转化条件为:初始pH 8.0,两步活化法以4%的接种量转接,48 h后加入底物(以N,N-二甲基甲酰胺溶解),发酵液中去氢骆驼蓬碱终浓度为0.25 mmoL/L,发酵时间为5 d。转化条件优化后H1和H2的产率分别达到8.70%和12.76%;该研究丰富了S.griseus ATCC 13273的作用底物范围,为哈尔酚和N-羟基去氢骆驼蓬碱的合成提供了一条新途径。
Using the method of microbial transformation, we selected Streptomyces griseus ATCC 13273 as the transformed strain and studied the transformation of the harrahine. The strain can convert the harmala base to two transformation products (denoted as H1 and H2). The products of the transformation were identified as HH and N-hydroxyl dehydrogenase by MS, 1H-NMR and 13C-NMR analysis, respectively Pomine The optimum conditions for the transformation were as follows: initial pH 8.0, 4% inoculum transfer by two-step activation method and substrate addition after 48 h (dissolved in N, N-dimethylformamide ). The final concentration of dahurian in fermentation broth was 0.25 mmoL / L, and the fermentation time was 5 d. The yield of H1 and H2 reached 8.70% and 12.76% respectively after optimization of transformation conditions. This study enriched the substrate range of S. griseus ATCC 13273, provided the synthesis of Halbin and N-hydroxyl harmine A new way.
