目的:研究细胞毒类化疗药——顺铂诱导人颊癌细胞凋亡发生的规律以及凋亡分子Apo-1的表达。探讨口腔癌化疗的细胞凋亡机制。方法:以建株人颊癌细胞系BcaCD885为对象,采用光镜、电镜以及流式细胞术等手段观察顺铂诱导颊癌细胞凋亡的形态特征及规律,同时检测顺铂对颊癌细胞Apo-1表达的影响。结果:顺铂作用后的颊癌细胞出现典型的凋亡细胞形态学特征;顺铂诱导颊癌细胞凋亡呈现时间及剂量依赖性效应关系;顺铂能上调颊癌细胞Apo-1的表达。结论:细胞凋亡可能是顺铂杀灭颊癌细胞的重要机制,Apo-1在顺铂诱导颊癌细胞凋亡中可能起着关键性调节作用
OBJECTIVE: To study the regularity of apoptosis induced by cisplatin and the expression of apoptosis-related protein Apo-1 in cytotoxic chemotherapy drug cisplatin. To investigate the apoptosis mechanism of oral cancer chemotherapy. Methods: The morphological characteristics and regularity of apoptosis induced by cisplatin in buccal carcinoma cells were observed by light microscopy, electron microscopy and flow cytometry. The apoptosis rate of buccal carcinoma cells Apo -1 expression. Results: The morphological features of typical apoptotic cells were observed in cisplatin-treated buccal carcinoma cells. The apoptosis of buccal carcinoma cells induced by cisplatin showed a time-and dose-dependent effect. Cisplatin could up-regulate the expression of Apo-1 in buccal carcinoma cells. CONCLUSION: Apoptosis may be an important mechanism of cisplatin in killing buccal carcinoma cells. Apo-1 may play a key regulatory role in cisplatin-induced apoptosis of buccal carcinoma cells
