目的:观察具有益气养阴活血作用的丹蛭降糖胶囊(DJC)对糖尿病大鼠胰腺炎症小体NLRP3的影响。方法:取大鼠55只,除正常对照组9只外,其余大鼠高能量饮食4周后加小剂量一次性腹腔注射STZ的方法建立2型糖尿病大鼠模型;造模成功后,模型组大鼠分为模型组,DJC高、低剂量组和吡格列酮组。各药物组分别予DJC(1.08、.0.54 g·kg-1·d-1)、吡格列酮10 mg·kg-1·d-1,正常组、模型组予等量生理盐水灌胃干预,连续8周。检测大鼠血糖及血脂水平;实时荧光定量PCR方法检测大鼠胰腺组织NLRP3mRNA表达。结果:与正常对照组比较,模型组胰腺炎症小体NLRP3 mRNA表达明显升高(P<0.01)。与模型组比较,DJC高剂量组NLRP3 mRNA表达明显降低(P<0.01);DJC高、低剂量组及吡格列酮组的血糖和血脂水平显著降低(P<0.01),而各用药组之间比较无明显差异。结论:DJC可控制NLRP3的过度表达,抑制细胞炎症反应、改善微炎症状态,从而减轻氧化应激损伤、保护胰岛B细胞。
Objective: To observe the effect of danzhijiangtang capsule (DJC) with Qi, nourishing Yin and promoting blood circulation on the expression of NLRP3 in diabetic rat pancreatitis body. Methods: 55 rats were randomly divided into normal control group (n = 9) and high-energy diet for 4 weeks, and then a small dose of intraperitoneal injection of STZ was used to establish type 2 diabetic rat model. Compared with the model group, the expression of NLRP3 mRNA in high-dose DJC group was significantly decreased (P <0.01), and the levels of blood glucose and blood lipid in high-dose, low-dose and high-dose pioglitazone groups were significantly lower (P <0.01) Significant differences. CONCLUSION: DJC can control the over-expression of NLRP3, inhibit the inflammatory reaction of cells and improve the micro-inflammatory state, so as to reduce the oxidative stress injury and protect islet B cells.
