目的 观察雷公藤多甙(GTW)对体外培养成骨细胞(OB)增殖的影响,以进一步探讨雷公藤(TW)致女性骨质疏松的机制。方法 以SD大鼠OB增殖为模型,分离新生SD大鼠头盖骨OB,传代后加入不同浓度GTW,用MTT比色分析法检测GTW在体外对OB增殖的作用和效价。结果 培养2d,GTW1μg/ml起显著抑制OB增殖(P<0.01);培养4d,GTW0.1μg/ml起显著抑制OB增殖(P<0.05)。药物对OB增殖的抑制程度与其剂量呈正相关(P<0.05)。结论 GTW显著抑制OB增殖且呈剂量依赖性。TW对OB的直接抑制作用,是其导致药物性骨质疏松的一个重要原因。
Objective To observe the effect of tripterygium wilfordii glycosides (GTW) on the proliferation of cultured osteoblasts (OB) in vitro and to explore the mechanism of female osteoporosis caused by TW. Methods The OB of SD rats was isolated from adult OB rat models. After passage, different concentration of GTW was added. The effect of GTW on OB proliferation was evaluated by MTT colorimetric assay. Results After cultured for 2 days, GTW1μg / ml significantly inhibited OB proliferation (P <0.01). After cultured for 4 days, GTW 0.1μg / ml significantly inhibited OB proliferation (P <0.05). The inhibitory effect of drugs on OB proliferation was positively correlated with its dosage (P <0.05). Conclusion GTW significantly inhibits OB proliferation in a dose-dependent manner. The direct effect of TW on the inhibition of OB is an important reason for the drug-induced osteoporosis.
