目的:探讨加味关节康对佐剂性关节炎大鼠免疫性炎症的影响。方法:53只大鼠随机分为4组:正常组、模型组、加味关节康剂治疗组及雷公藤多甙片组,除正常组外其余大鼠制备佐剂性关节炎模型。治疗组分别用加味关节康、雷公藤多甙片ig;于药后1,2,3周测大鼠左后足趾肿胀度及关节炎指数评分。用药后3周测定血清白介素(IL)-4、8、10、12,肿瘤坏死因子(TNF)-α和干扰素(IFN-γ)水平,观察大鼠滑膜组织病理形态学变化。结果:用药后治疗组关节肿胀度及关节炎指数均小于模型组(P<0.05,P<0.01);用药3周后各治疗组血清TNF-α、IFN-γ、IL-8、IL-12水平均低于模型组(P<0.01);IL-4、IL-10水平均高于模型组(P<0.05);各治疗组与模型组比较滑膜组织增生程度明显减轻,炎症细胞浸润程度稍减轻。结论:加味关节康治疗可明显下调佐剂性关节炎大鼠相关致炎性细胞因子TNF-α、IFN-γ、IL-8、IL-12水平,并上调抗炎性细胞因子IL-4、IL-10的水平,可减轻滑膜增生程度,改善关节肿胀,疗效肯定。
Objective: To investigate the effect of Jiajiejiekang on immune inflammation in adjuvant arthritis rats. METHODS: Fifty-three rats were randomly divided into 4 groups: normal group, model group, Jiawei Synesthesia group and Tripterygium wilfordii group. In addition to the normal group, adjuvant arthritis models were prepared. The rats in the treatment group were treated with Jiagen Jiekang and Tripterygium wilfordii ig respectively. The degree of swollenness of the left hind toe and arthritis index scores were measured at 1, 2 and 3 weeks after the treatment. Serum interleukin (IL)-4, 8, 10, 12, tumor necrosis factor (TNF)-α and interferon (IFN-γ) levels were measured at 3 weeks after administration. Pathological changes in synovial tissue were observed. Results: The degree of joint swelling and arthritis index in the treatment group after treatment were smaller than those in the model group (P<0.05, P<0.01); serum TNF-α, IFN-γ, IL-8, and IL-12 in the treatment groups after 3 weeks of treatment The average blood levels were lower than those in the model group (P<0.01); the levels of IL-4 and IL-10 were higher than those in the model group (P<0.05); the degree of synovial tissue proliferation was significantly reduced and the degree of inflammatory cell infiltration was significantly reduced in each treatment group compared with the model group. Slightly reduced. Conclusion: Jiajie Jiekang treatment can significantly reduce the levels of TNF-α, IFN-γ, IL-8 and IL-12, and upregulate anti-inflammatory cytokine IL-4 in adjuvant-induced arthritis rats. The level of IL-10 can reduce the degree of synovial hyperplasia and improve the swelling of joints.
