目的:探讨二乙酰二脱水卫矛醇(diacetyldianhydrogalactitol,DADAG)对小鼠白血病L1210细胞凋亡相关蛋白表达的影响。方法:MTT法测定DADAG对L1210细胞的杀伤作用;流式细胞技术和透射电镜检测细胞凋亡;Western blot法分析细胞内Bcl-2和Bcl-X_L蛋白水平的变化。结果:与对照组相比,DADAG可降低L1210细胞的存活率,且具有浓度依赖性。给药12.0、17.2、24.5、35.0和50.0mg/L 24h后,流式细胞术周期分析结果显示,各处理组均出现凋亡峰。DADAG 50.0mg/L作用L1210细胞24h后,靶细胞出现固缩、染色质边集;DADAG可下调Bcl-2和Bcl-X_L蛋白水平,且具有时间依赖性。结论:DADAG可能通过抑制Bcl-2及Bcl-X_L蛋白的表达而诱导L1210细胞发生凋亡。
Objective: To investigate the effect of diacetyldianhydrogalactitol (DADAG) on the expression of apoptosis-related proteins in mouse leukemia L1210 cells. Methods: The killing effect of DADAG on L1210 cells was determined by MTT assay. The apoptosis of L1210 cells was detected by flow cytometry and transmission electron microscopy. The changes of Bcl-2 and Bcl-X_L protein were analyzed by Western blot. Results: Compared with the control group, DADAG can reduce the survival rate of L1210 cells in a concentration-dependent manner. After administration of 12.0, 17.2, 24.5, 35.0 and 50.0 mg / L for 24 h, the results of flow cytometry analysis showed that apoptotic peak appeared in all treatment groups. After treated with DADAG 50.0 mg / L for 24 h, the target cells were condensed and chromatin margined. DADAG down-regulated Bcl-2 and Bcl-X_L protein levels in a time-dependent manner. Conclusion: DADAG may induce the apoptosis of L1210 cells by inhibiting the expression of Bcl-2 and Bcl-X_L proteins.
