[目的]探讨低强度微波辐射对盐酸阿霉素(DOX)致人早幼粒白血病HL-60细胞损伤的影响。[方法]将HL-60细胞随机分为对照组、单纯微波组、单纯DOX组和联合组(微波+DOX)。单纯微波组和联合组都给予12μW/cm2微波照射,1h/d,连续辐照3d,对照组和单纯DOX组置于同一环境,但不给予电磁辐射。第4天分别给予单纯DOX组和联合组以浓度为0.125mg/L的DOX。用钙离子依赖性磷脂结合蛋白-异硫氰酸荧光素/碘化丙啶(Annexin V-FITC/PI)检测细胞凋亡率,流式细胞仪检测线粒体膜电位和胞内游离Ca2+浓度。[结果]与对照组相比,单纯DOX组凋亡率明显增加(P<0.05);线粒体膜电位明显下降(P<0.05);胞内游离Ca2+浓度明显升高(P<0.05)。与单纯DOX组相比,联合组凋亡率明显降低(P<0.05);线粒体膜电位明显升高(P<0.05);游离Ca2+浓度明显降低(P<0.05)。[结论]预先900MHz、12μW/cm2低强度微波照射能够明显减轻DOX引起的细胞损伤。
[Objective] To investigate the effect of low-intensity microwave radiation on the injury of human promyelocytic leukemia HL-60 cells induced by doxorubicin hydrochloride (DOX). [Methods] HL-60 cells were randomly divided into control group, simple microwave group, simple DOX group and combined group (microwave + DOX). Microwave group and combination group were given 12μW / cm2 microwave irradiation, 1h / d, continuous irradiation 3d, control group and pure DOX group placed in the same environment, but did not give electromagnetic radiation. Day 4 were given DOX group and the combination group to a concentration of 0.125mg / L of DOX. The rate of apoptosis was detected by calcium ion-dependent phospholipid-binding protein-fluorescein isothiocyanate / propidium iodide (PI) and the mitochondrial membrane potential and intracellular free Ca2 + concentration were detected by flow cytometry. [Results] Compared with the control group, the apoptotic rate of DOX group was significantly increased (P <0.05); mitochondrial membrane potential was significantly decreased (P <0.05); intracellular free Ca2 + concentration was significantly increased (P <0.05). Compared with DOX alone group, the apoptotic rate in the combined group was significantly decreased (P <0.05), the mitochondrial membrane potential was significantly increased (P <0.05), and the free Ca2 + concentration was significantly decreased (P <0.05). [Conclusion] The pre-900MHz, 12μW / cm2 low-intensity microwave irradiation can significantly reduce DOX-induced cell damage.
