为了解原发性皮肤T细胞淋巴瘤(PCTCL)外周血中克隆性T细胞的存在及其与临床疗效的相关性,分别对25例PCTCL患者经重组α干扰素治疗前后的外周血标本,应用聚合酶链反应(PCR)方法,以通用引物扩增T细胞受体(TCR)γ链编码基因中V(可变区)J(连接区)的结合序列,检测代表克隆性T细胞分子标志的TCR基因重排(GR)。结果发现,25例PCTCL外周血样品中,17例见克隆性T细胞(示TCRGR),经有效治疗后减至3例,治疗前后克隆性T细胞的检出率具显著性差异。说明克隆性T细胞在PCTCL外周血中常见,外周血中克隆性T细胞的动态监测对PCTCL临床疗效的判定有辅助作用。
To understand the presence of clonal T cells in peripheral blood of primary cutaneous T-cell lymphoma (PCTCL) and its correlation with clinical efficacy, peripheral blood samples before and after treatment with recombinant interferon alpha in 25 PCTCL patients were used. Polymerase chain reaction (PCR) method using universal primers to amplify the V (variable region)-J (binding region) binding sequence in the T-chain receptor (TCR) gamma chain-encoding gene to detect molecular signatures representing clonal T-cells TCR gene rearrangement (GR). The results showed that in 25 cases of PCTCL peripheral blood samples, 17 cases saw clonal T cells (TCGRR), after effective treatment was reduced to 3 cases, the detection rate of clonal T cells before and after treatment had significant differences. It shows that clonal T cells are common in peripheral blood of PCTCL, and the dynamic monitoring of clonal T cells in peripheral blood can help determine the clinical efficacy of PCTCL.
