目的:探讨宁心红杞胶囊对围绝经大鼠卵巢颗粒细胞凋亡及其相关基因Bax和Bc l-2表达的影响。方法:选用4月龄雌性大鼠为正常对照组,14月龄阴道细胞学表现动情期延长的雌性大鼠作为衰老大鼠模型。模型动物随机分别给宁心红杞胶囊或补佳乐治疗。采用原位末端核苷酸标记法(TUNEL)检测卵巢细胞凋亡,免疫组化法检测凋亡相关因子Bc l-2、Bax。结果:宁心红杞胶囊高、中剂量组卵巢细胞凋亡阳性表达与模型组比较有显著性差异,同时可使卵巢抑凋亡基因Bc l-2表达增强,促凋亡基因Bax表达减弱,Bc l-2/Bax比例增加。结论:宁心红杞胶囊通过抑制Bax蛋白的表达,上调Bc l-2蛋白表达,延缓卵巢颗粒细胞凋亡,进而改善卵巢功能。
Objective: To investigate the effect of Ningxin Hongqi Capsule on apoptosis of ovarian granulosa cells and expression of related genes Bax and Bcl-2 in perimenopause rats. METHODS: Four-month-old female rats were used as the normal control group. Female rats aged 14 months of vaginal cytology with prolonged estrus were used as the aging rat model. Model animals were randomly given to Ningxin Hongqi Capsule or Jiajiale for treatment. Apoptosis of ovarian epithelial cells was detected by TUNEL. The apoptosis-related factors Bcl-2 and Bax were detected by immunohistochemistry. RESULTS: The positive expression of apoptosis in ovarian cancer cells in the high and middle dose group of Ningxin Hongqi Capsule was significantly different from that in the model group. At the same time, the ovarian suppressor gene Bcl-2 expression was enhanced and the proapoptotic gene Bax expression was decreased. The proportion of Bc l-2/Bax increased. Conclusion: Ningxin Hongqi Capsule can improve the ovarian function by inhibiting the expression of Bax protein, up-regulating the expression of Bcl-2 protein, and delaying the apoptosis of ovarian granulosa cells.
