目的:通过多指标综合评分法优选小儿清肺止咳巴布膏醇提工艺,确定最佳工艺参数。方法:以处方工艺中麻黄碱、隐丹参酮的提取率及浸出物含量为指标,通过多指标综合评分法,考察醇浓度、溶剂倍量、提取时间及提取次数对小儿清肺止咳巴布膏醇提工艺的影响。采用HPLC法测定麻黄碱和隐丹参酮的含量,流动相分别为乙腈-0.1%磷酸水溶液(5∶95)和甲醇-水(77∶23),检测波长分别为210和260 nm。结果:通过正交实验优选的最佳提取工艺:加15倍量70%乙醇提取2次,第1次加8倍量溶剂提取2 h,第2次加7倍量溶剂提取1 h。麻黄碱和隐丹参酮的提取率及浸出物得率分别为74.44%,85.23%和39.57%。结论:采用多指标综合评分法优选的的醇提工艺科学合理,有效成分提取率高。
OBJECTIVE: To optimize the optimal extraction process of Qingfeizhibabu cream by multi-index comprehensive score method. Methods: Taking the extraction rate of ephedrine and the content of cryptotanshinone in the prescription process as the index, the alcohol concentration, the solvent doubling time, the extraction time and the number of extraction were investigated by multi-index comprehensive score method. Mention the impact of technology. The contents of ephedrine and cryptotanshinone were determined by HPLC. The mobile phases were acetonitrile-0.1% phosphoric acid solution (5:95) and methanol-water (77:23) respectively. The detection wavelength was 210 and 260 nm, respectively. Results: The optimal extraction process was optimized by orthogonal experiment: two times with 15 times the amount of 70% ethanol, the first with 8 times the amount of solvent extracted 2 h, the second with 7 times the amount of solvent extraction 1 h. The extraction rate of ephedrine and cryptotanshinone and the yield of extract were 74.44%, 85.23% and 39.57% respectively. Conclusion: The optimized alcohol extraction process by using multi-index comprehensive scoring method is scientific and reasonable, and the extraction rate of active ingredients is high.
