目的依据生育三烯酚的抗肿瘤作用,研究其对人结肠癌细胞SW620细胞增殖抑制作用与Wnt信号途径之间的关系。方法采用MTT(噻唑蓝比色法)方法,观察不同剂量δ-生育三烯酚对细胞增殖抑制作用,通过免疫细胞化学染色法和蛋白免疫印迹方法测定Wnt信号途径中相关因子的蛋白表达情况。结果δ-生育三烯酚对结肠癌SW620细胞增殖有明显抑制作用,20μmol/Lδ-生育三烯酚作用24h对SW620细胞抑制率为70.43%,IC50值为15.18μmol/L。20μmol/Lδ-生育三烯酚可使人结肠癌细胞SW620中Wnt信号途径中的wnt-1、β-catenin、c-jun和cylin D1蛋白表达量显著下降(P<0.05)。结论δ-生育三烯酚对结肠癌SW620细胞增殖抑制作用可能与其致Wnt途径中wnt-1、β-catenin、c-jun和cylin D1表达降低有关。
Objective To study the anti-tumor effect of tocotrienol on the proliferation of human colon cancer cell line SW620 and its relationship with Wnt signaling pathway. Methods MTT assay was used to observe the inhibitory effect of different doses of δ-tocotrienol on cell proliferation. Immunocytochemical staining and Western blotting were used to detect the protein expression of related factors in Wnt signaling pathway. Results δ-Tocotrienol had a significant inhibitory effect on the proliferation of SW620 cells. The inhibitory rate of SW620 cells treated with 20μmol / L δ-Tocotrienol for 24 hours was 70.43% and the IC50 was 15.18μmol / L. The expression of wnt-1, β-catenin, c-jun and cylin D1 in Wnt signaling pathway of human colon cancer cell SW620 was significantly decreased by 20μmol / L δ-tocotrienol (P <0.05). Conclusion The inhibitory effect of δ-tocotrienol on the proliferation of colon cancer SW620 cells may be related to the decrease of Wnt-1, β-catenin, c-jun and cylin D1 expression in Wnt pathway.
