目的:探讨低分子量姬松茸多糖(LMPAB)对肿瘤细胞与内皮细胞侵袭和黏附的影响和作用。方法:不同剂量的LMPAB(5,10,20 mg.L-1)分别作用于人结肠癌细胞(HT-29)和人脐静脉内皮细胞(HUVECs)48 h,采用MTT法检测LMPAB对HT-29和HUVECs增殖的影响、采用分光光度法检测LMPAB对HT-29细胞与HUVECs黏附的影响、采用Transwell双层小室观察LPMPAB对HT-29迁移穿过单层HUVECs的影响。结果:不同剂量LMPAB(5,10,20 mg.L-1)对HT-29和HUVECs增殖无显著性作用;使HT-29黏附于单层HUVECs的数量得到了明显的抑制,抑制率分别为26.96%,23.60%,27.66%;对HT-29迁移穿过HUVECs也具有抑制作用,抑制率分别为23.96%,23.59%,41.36%。结论:低分子量姬松茸多糖通过抑制HT-29肿瘤细胞与内皮细胞的黏附以及肿瘤细胞迁移穿过内皮细胞而发挥其抗肿瘤转移作用。
Objective: To investigate the effect of low molecular weight Agaricus blazei polysaccharide (LMPAB) on the invasion and adhesion of tumor cells and endothelial cells. Methods: Different concentrations of LMPAB (5, 10, 20 mg.L-1) were administered to human colon cancer cells (HT-29) and human umbilical vein endothelial cells (HUVECs) for 48 h. MTT assay was used to detect the effect of LMPAB on HT- 29 and HUVECs. The effect of LMPAB on the adhesion of HT-29 cells to HUVECs was detected by spectrophotometry. The effect of LPMPAB on the migration of HT-29 through HUVECs was observed by Transwell double chamber. Results: LMPAB (5,10,20 mg.L-1) had no significant effect on the proliferation of HT-29 and HUVECs. The number of HT-29 adhered to the monolayer of HUVECs was significantly inhibited 26.96%, 23.60% and 27.66%, respectively. HT-29 also inhibited the migration of HUVECs with the inhibitory rates of 23.96%, 23.59% and 41.36%, respectively. Conclusion: The low molecular weight Agaricus blazei polysaccharides exert its antitumor effect by inhibiting the adhesion of HT-29 tumor cells to endothelial cells and the migration of tumor cells through endothelial cells.
