目的 观察基因修饰的骨髓基质细胞对辐射的耐受作用。方法 通过辐射对CFU F形成的影响制作细胞存活曲线分析造血因子双顺反子基因修饰的基质细胞 (HFCL FG和HFCL EFG)对辐射的耐受 ,采用3H TdR掺入法验证上述结果 ,观察转染细胞对粒系祖细胞生长的影响。结果 HFCL FG和HFCL EFG细胞生存曲线的肩明显增宽 ,D0 值增大 ,N和Dq值增大 ,3H TdR掺入实验也验证了该结果。这种细胞对粒系祖细胞的扩增作用明显增强。结论 造血因子基因修饰的骨髓基质细胞具有抗辐射和支持造血作用。
Objective To observe the radiation tolerance of genetically modified bone marrow stromal cells. Methods The effect of radiation on CFU F formation Cell survival curve was used to analyze the radiation tolerance of the bi-cistronic gene modified stromal cells (HFCL FG and HFCL EFG). The 3H-TdR incorporation assay was used to verify the above results. Effect of Dye Cells on the Growth of Granulocyte Progenitor Cells. Results The shoulder of survival curves of HFCL FG and HFCL EFG cells were obviously broadened, D0 values increased, N and Dq values increased, and 3H TdR incorporation experiments also verified the results. The proliferation of myeloid cells in this kind of cells is obviously enhanced. Conclusion Hematopoietic gene-modified bone marrow stromal cells have anti-radiation and hematopoietic effects.
