目的从罗氏海盘车(Asterias rollestoni)中分离鉴定生物碱类天然产物,并进行体外抗肿瘤活性评价。方法利用凝胶吸附树脂柱层析、硅胶薄层层析、硅胶柱层析、凝胶Sephadex LH-20柱层析和高效液相色谱等手段对化合物进行了分离纯化;运用NMR、MS等波谱方法并结合文献鉴定化合物的结构;采用SRB、MTT法评价化合物的体外抗肿瘤活性。结果从罗氏海盘车的乙醇提取物中分离鉴定了1个吲哚生物碱N-(2-苯乙基)-1H-吲哚-3-乙胺(1)和1个环二肽生物碱类化合物Fellutanine A(2)。化合物2对MGC803细胞的抑制率为53.99%,具有一定的抗肿瘤活性。结论 2个生物碱类化合物均为首次从罗氏海盘车中分离获得;化合物1为首次分离到的天然产物,其核磁数据为首次报道;化合物2具有一定的抗肿瘤活性。
Objective To isolate and identify natural alkaloids from Asterias rollestoni and evaluate the antitumor activity in vitro. Methods The compounds were isolated and purified by gel filtration resin column chromatography, silica gel thin layer chromatography, silica gel column chromatography, gel Sephadex LH-20 column chromatography and high performance liquid chromatography. By NMR, MS and other spectroscopy Methods The compounds were identified by the literature. The anti-tumor activity of the compounds was evaluated by SRB and MTT. Results One indole alkaloid N- (2-phenylethyl) -1H-indole-3-ethylamine (1) and one cyclic dipeptide alkaloid Compound Fellutanine A (2). The inhibitory rate of compound 2 to MGC803 cells was 53.99%, which showed some antitumor activity. Conclusions Both alkaloids are isolated from Roche Haeundae for the first time. Compound 1 is the first natural product isolated and its first NMR data are reported. Compound 2 has some antitumor activity.
