目的对进口黑虎虾中分离的菌株F14-2进行种属鉴定,通过顶空气相色谱-质谱联用法(headspace gas chromatography-mass spectrometry,HS-GC-MS)分析分离菌株液态培养代谢的挥发性产物。方法应用VITEK 2 Compact全自动微生物鉴定系统分析菌株的生理生化特征,利用荧光定量PCR特异性扩增检测病原菌,通过顶空气相色谱-质谱联用法分析分离菌株F14-2的代谢的挥发性产物。结果菌株F14-2为革兰氏阴性菌,生理生化特征与霍乱弧菌(Vibrio cholerae)的相似性为98%,荧光定量PCR检测和血清凝集试验进一步确认为非O1非O139群霍乱弧菌,菌株F14-2在营养肉汤(nutrient broth)中代谢的挥发性产物主要有乙醇、异戊醇、乙酸和2-乙基己醇。结论进口黑虎虾中分离的菌株F14-2鉴定为霍乱弧菌,该菌产生的挥发性产物为霍乱弧菌的鉴定提供参考。
Objective To identify the strain F14-2 isolated from imported black tiger shrimp and analyze its volatile components by liquid chromatography-headspace gas chromatography-mass spectrometry (HS-GC-MS) product. Methods The physiological and biochemical characteristics of the strain were analyzed by VITEK 2 Compact automated microbial identification system. Pathogenic bacteria were detected by fluorescence quantitative PCR. The metabolites of the strain F14-2 were analyzed by headspace gas chromatography - mass spectrometry. Results The strain F14-2 was a Gram-negative bacterium. The similarity of physiological and biochemical characteristics with Vibrio cholerae was 98%. Fluorescent quantitative PCR and serum agglutination test further confirmed that non-O1 non-O139 Vibrio cholerae, The volatile products metabolized by strain F14-2 in nutrient broth are mainly ethanol, isoamyl alcohol, acetic acid and 2-ethylhexanol. Conclusion The strain F14-2 isolated from imported black tiger shrimp was identified as Vibrio cholerae, and the volatile product produced by this strain was a reference for the identification of V. cholerae.
