目的探讨益气活血复方芪丹通脉片(QDTMT)对缺血/再灌注(I/R)大鼠心肌中核因子-κB(NF-κB)表达的影响。方法将SD大鼠48只随机分为6组(n=8),即假手术组、模型组、QDTMT低中高剂量组(0.36、1.08、3.24g/kg)及恬尔心组(5 mg/kg)。各组均用生理盐水配制的等体积药液灌胃7 d,每日1次。采用冠脉结扎的方法,造成大鼠心肌I/R模型后,采用Western blot和免疫组化染色法,检测各组大鼠心肌中NF-κB蛋白的表达。结果与假手术组比较,模型组NF-κB的表达显著增多(P<0.01)。与模型组比较,QDTMT各剂量组和恬尔心组NF-κB的表达显著减少(P<0.01)。结论QDTMT能抑制I/R大鼠心肌中NF-κB蛋白的表达,从而对心肌具有保护作用。
Objective To investigate the effect of QDTMT on the expression of nuclear factor-κB (NF-κB) in rat myocardium during ischemia/reperfusion (I/R). Methods Forty-eight Sprague-Dawley rats were randomly divided into 6 groups (n=8), namely sham-operated group, model group, QDTMT low-medium high-dose group (0.36, 1.08, 3.24g/kg) and Dierxin group (5mg/kg). Kg). Each group was intragastrically administrated with an equal volume of saline for 7 days, once daily. After myocardial I/R model was established by coronary artery ligation, the expression of NF-κB protein in rat myocardium was detected by Western blot and immunohistochemical staining. Results Compared with the sham group, the expression of NF-κB in the model group increased significantly (P<0.01). Compared with the model group, the expression of NF-κB was significantly decreased in each QDTMT dose group and Dura Heart group (P<0.01). Conclusion QDTMT can inhibit the expression of NF-κB protein in the myocardium of I/R rats, which has a protective effect on myocardium.
