目的探讨人子宫颈鳞癌发生发展过程中,膜联蛋白A5(ANXA5)表达的变化。方法收集人子宫颈鳞癌组织25例和人正常子宫颈组织15例,细胞裂解液裂解组织细胞,蛋白定量试剂盒进行蛋白定量后,Western blotting法检测ANXA5在人子宫颈鳞癌组织和人正常子宫颈组织中的表达;为进一步确定ANXA5的表达部位及其变化,我们收集人子宫颈鳞癌蜡块45例和人正常子宫颈组织蜡块20例,用免疫组织化学ABC法对两种组织ANXA5的表达进行检测,原位杂交法在RNA水平进行检测。结果Westen blotting显示人子宫颈鳞癌组织ANXA5比人正常子宫颈组织表达增强;免疫组织化学及原位杂交均显示人子宫颈癌组织中ANXA5的染色比人正常子宫颈组织染色重,阳性信号分布于细胞质和细胞膜。结论ANXA5在人子宫颈鳞癌组织中的表达比在人正常子宫颈组织中的表达增高;ANXA5与人子宫颈鳞癌的发生有密切联系。
Objective To investigate the expression of annexin A5 (ANXA5) in the development and progression of human cervical squamous cell carcinoma. Methods Twenty-five human cervical squamous cell carcinoma tissues and 15 normal human cervical tissues were collected. The cell lysate was lysed and the protein quantitation kit was used for protein quantification. Western blotting was used to detect the expression of ANXA5 in human cervical squamous cell carcinoma and normal human Cervical tissue. To further determine the expression site of ANXA5 and its changes, we collected 45 cases of cervical squamous cell carcinoma of the cervix and 20 cases of normal human cervical tissue wax blocks. Immunohistochemical ABC method was used to detect the expression of both tissues ANXA5 expression was detected in situ hybridization at the RNA level for testing. Results Westen blotting showed that human cervical squamous cell carcinoma ANXA5 expression than normal human cervical tissue enhanced; immunohistochemistry and in situ hybridization showed human cervical cancer tissue ANXA5 staining than normal human cervical tissue staining, positive signal distribution In the cytoplasm and cell membrane. Conclusions The expression of ANXA5 in human cervical squamous cell carcinoma is higher than that in human normal cervical tissue. ANXA5 is closely related to the occurrence of cervical squamous cell carcinoma in human.
