随着生物技术的迅速发展,许多肽类药物已用于临床治疗各种疾病。为了避免注射,人们已经研究了许多给药途径,其中眼部给药似乎是最可行的一种途径。因为(1)与注射一样,眼部给药剂量准确;(2)与注射相比眼部给药显得非常容易、经济;(3)全身吸收速率与注射同样迅速;(4)眼组织对发生免疫反应的敏感性比其它组织低得多;(5)通过眼部吸收的药物可以避免肝脏的首过代谢或分解;(6)长期(3个月)每天使用胰岛素滴眼液没有产生耐受性或其它眼科不良反应。
With the rapid development of biotechnology, many peptide drugs have been used clinically to treat various diseases. To avoid injection, many routes of administration have been investigated, of which ophthalmic administration seems to be the most viable route. Because (1) the same amount of ophthalmic dose administered as injections; (2) ophthalmic administration is considerably easier and less costly than injections; (3) systemic absorption rates are as rapid as injections; (4) Immune response is much less sensitive than other tissues; (5) First-pass metabolism or decomposition of the liver can be avoided by ophthalmically absorbed drugs; (6) Insulin drops are not tolerated daily by long-term (3 months) Sexual or other ophthalmic adverse reactions.
