目的观察细胞因子对蜕膜细胞介导B淋巴细胞分泌IgG的影响,探讨蜕膜局部免疫微环境的免疫学特点。方法将γ-干扰素(IFN-γ)、白细胞介素-2(IL-2)、白细胞介素-6(IL-6)和表皮生长因子(EGF)加入培养的蜕膜细胞中,用蜕膜细胞的培养上清液刺激B淋巴细胞生长,通过放射免疫法测定作用不同时间后B细胞分泌的IgG的值。结果正常早孕蜕膜细胞的培养上清液可以刺激B淋巴细胞IgG的分泌;经细胞因子作用后的蜕膜细胞培养上清液使B细胞分泌的IgG明显上升,但这种作用与细胞因子的浓度、种类无明显相关性,而与细胞因子作用的时间具有一定关联,表现在第12小时实验组的IgG分泌显著上升(P<0.05),到24、48h时实验组有逐渐下降趋势。结论在外源性细胞因子的作用下,蜕膜局部体液免疫反应增强,但这种增强可能是蜕膜局部免疫的自身调节作用,以维护妊娠的正常进行。
Objective To observe the effect of cytokines on the secretion of IgG by B lymphocytes induced by decidual cells and to investigate the immunological characteristics of decidual immunoenvironment. Methods IFN-γ, IL-2, IL-6 and epidermal growth factor (EGF) were added to cultured decidual cells, The culture supernatant of membrane cells stimulated the growth of B lymphocytes, and the value of IgG secreted by B cells at different time after the action was determined by radioimmunoassay. Results The culture supernatant of normal early decidual cells stimulated the secretion of IgG from B lymphocytes. The supernatant of decidual cells after cytokines stimulated the secretion of IgG from B cells. However, this effect was not associated with cytokines There was no significant correlation between concentration and type, but correlated with the time of cytokines. The IgG secretion in experimental group increased significantly at 12 hours (P <0.05), and gradually decreased at 24 and 48 hours. Conclusion The excretion of humoral immune response in decidua is enhanced by exogenous cytokines, but this enhancement may be the self-regulation of decidual local immunity to maintain the normal pregnancy.
