目的:使用已构建的真核表达载体pEGFP-CLL1转染中国仓鼠卵巢(CHO)细胞,探讨CL-L1在CHO细胞内的定位。方法:使用免疫荧光标记技术和Western blot检测EGFP-CLL1融合蛋白在细胞内的表达部位。结果:融合蛋白主要存在于细胞质基质中,不存在于内质网、高尔基体和细胞核等部位。结论:CL-L1为已发现惟一存在于细胞质基质中的胶原样凝集素成员。
OBJECTIVE: To transfect Chinese hamster ovary (CHO) cells with the constructed eukaryotic expression vector pEGFP-CLL1 to investigate the localization of CL-L1 in CHO cells. Methods: Immunofluorescent labeling and Western blot were used to detect the expression of EGFP-CLL1 fusion protein in cells. Results: The fusion protein mainly exists in the cytoplasm, but not in the endoplasmic reticulum, Golgi and nucleus. CONCLUSION: CL-L1 is a collagen-like lectin that has been found exclusively in the cytoplasm matrix.
