目的:应用银芍散干预高脂饮食诱导的单纯性脂肪肝模型大鼠,明确银芍散对非酒精性单纯性脂肪肝的防治作用。方法:采用高脂饲料加10%果糖水喂养18周,诱导SD大鼠形成单纯性脂肪肝,造模同时灌胃给药,记录体重,观察一般行为学。末次给药后取下腔静脉血和肝脏,检测血清血脂四项(TC、TG、HDL-c、LDLc)及肝功能指标(ALT、AST)。计算肝指数,HE染色观察肝脏组织病理的变化,并检测肝组织中总脂酶活性和FFA水平。结果:与正常组相比,模型组大鼠体重增长较为明显,血清TC、TG、LDL-c、ALT、AST、肝指数及肝中FFA水平明显升高,血清HDL-C水平及肝总脂酶活性明显降低(P<0.01,P<0.05)。病理学显示模型组大鼠出现了严重脂肪变和不同程度炎症细胞浸润、汇管区渗出、肝细胞坏死。应用银芍散干预后,大鼠血清TC、TG、LDL-c、ALT、AST、肝指数及肝中FFA水平明显降低,血清HDL-C及肝总脂酶活性明显升高(P<0.01,P<0.05)。同时可明显减轻大鼠肝肿大,改善肝细胞的脂肪变性和炎症程度。结论:银芍散可通过纠正血脂紊乱,改善肝功能和肝细胞脂肪变性防治单纯性脂肪肝,其作用机制可能与提高肝总脂酶活性,减少肝中FFA,进而增加TG分解,降低TG合成有关。
OBJECTIVE: To study the preventive and therapeutic effects of Yin-Shao-San on non-alcoholic fatty liver in rats with simple fatty liver induced by high-fat diet. Methods: The rats were fed with high-fat diet plus 10% fructose and water for 18 weeks. SD rats were induced to form simple fatty liver. The models were given gavage at the same time. Body weight was recorded and general behavior was observed. After the last administration, the vena cava and liver were removed, and the serum lipids (TC, TG, HDL-c, LDLc) and liver function indexes (ALT, AST) were measured. The liver index was calculated, the histopathological changes of liver were observed by HE staining, and the total lipase activity and FFA level in liver tissue were detected. Results: Compared with the normal group, the body weight gain of the model group was more obvious. The levels of serum TC, TG, LDL-c, ALT, AST, liver index and FFA in the liver were significantly increased. The levels of serum HDL- The enzyme activity decreased significantly (P <0.01, P <0.05). Pathology showed that the rats in the model group developed severe steatosis and infiltration of inflammatory cells in different degrees, exudation in the portal area and necrosis of liver cells. The serum levels of TC, TG, LDL-c, ALT, AST, liver index and hepatic FFA in the rats were significantly decreased with the increase of the serum levels of HDL-C and total lipase (P <0.01, P <0.05). At the same time can significantly reduce rat hepatomegaly, to improve the degree of fatty liver degeneration and inflammation. Conclusion: Yinshuosan Powder can prevent and treat simple fatty liver by correcting dyslipidemia, improving liver function and fatty degeneration of liver cells. Its mechanism may be related to increasing hepatic total lipase activity, decreasing FFA in liver, increasing TG decomposition and decreasing TG synthesis related.
