目的:研究染料木素和青藤碱对佐剂型关节炎(AA)大鼠血清白介素17(IL-17)、抗酒石酸酸性磷酸酶(Tracp)含量的影响,以初步探讨其对AA大鼠骨免疫系统的调节作用。方法:通过给大鼠左后足跖皮下注射完全弗氏佐剂建立AA大鼠模型。注射诱导后第2天大鼠出现急性关节炎症状,踝关节不同程度肿胀,炎症持续第21天开始给予药物干预。染料木素组给予染料木素(1 mg/200 g.d-1)灌胃,青藤碱组给予青藤碱(3 mg/200 g.d-1)灌胃,配伍组给予染料木素(1 mg/200 g.d-1)和青藤碱(3 mg/200 g.d-1)灌胃,甲氨喋呤组给予甲氨喋呤(1 mg/200 g.w-1)灌胃。连续干预40d后用酶联免疫吸附法测定大鼠血清IL-17和Tracp的含量。结果:①配伍组与青藤碱组和甲氨喋呤组比较,IL-17、Tracp含量均明显降低(P<0.05,P<0.01);与染料木素组比较,IL-17、Tracp含量下调但差异不显著(P>0.05)。②染料木素组Tracp含量明显低于青藤碱组和甲氨喋呤组(P<0.05);染料木素组IL-17含量低于青藤碱组、高于甲氨喋呤组,但差异无统计学意义(P>0.05)。结论:染料木素和青藤碱合用能发挥协同作用,降低AA大鼠血清IL-17、Tracp含量。
Objective: To investigate the effects of genistein and sinomenine on the levels of serum interleukin 17 (IL-17) and tartrate-resistant acid phosphatase (Tracp) in adjuvant arthritis (AA) Modulation of the immune system. Methods: AA rat model was established by subcutaneous injection of complete Freund’s adjuvant into the left posterior plantar of rats. On the second day after injection, the rats showed symptoms of acute arthritis, varying degrees of ankle swelling, and the inflammation started on the 21st day to start the drug intervention. Genistein (1 mg / 200 gd-1) was administered intragastrically in the genistein group, sinomenine (3 mg / 200 gd-1) was administered to the sinomenine group, and genistein (1 mg / 200 gd-1) and sinomenine (3 mg / 200 gd-1) respectively. Methotrexate (1 mg / 200 gw-1) was given intragastrically in the methotrexate group. Serum levels of IL-17 and Tracp were measured by enzyme linked immunosorbent assay after continuous intervention for 40 days. Results: ① Compared with sinomenine group and methotrexate group, IL-17 and Tracp contents were significantly decreased (P <0.05, P <0.01); Compared with genistein group, IL-17 and Tracp contents But the difference was not significant (P> 0.05). ② The content of Tracp in genistein group was significantly lower than that in sinomenine group and methotrexate group (P <0.05). The content of IL-17 in genistein group was lower than that in sinomenine group and higher than that in methotrexate group The difference was not statistically significant (P> 0.05). Conclusion: The combination of genistein and sinomenine can exert a synergistic effect, reducing serum IL-17, Tracp content in AA rats.
