本实验用过氧化氢(H_2O_2)溶液作为外源性自由基,直接作用于正常兔脑组织表面诱导脑水肿。用电子自旋共振波谱仪(ESR)直接检测兔脑组织的自由基信号,同时检测了丙二醛(MDA)、脑组织水含量;并进行脑组织病理学观察。结果脑组织自由基、MDA、水含量都明显上升,和生理盐水对照组相比具有统计学差异,病理呈现混合型脑水肿改变。结果表明自由基及脂质过氧化是脑水肿形成和发展的重要分子病理机制之一;另外由于H_2O_2溶液能引起严重的脑水肿和损伤,因此临床上应避免使用。
In this experiment, hydrogen peroxide (H_2O_2) solution was used as an exogenous free radical to directly induce brain edema on the surface of normal rabbit brain tissue. Electron spin resonance spectroscopy (ESR) was used to directly detect the free radical signal of brain tissue in rabbits. Malondialdehyde (MDA) and brain tissue water content were detected at the same time. Brain pathological observation was also performed. Results The contents of free radicals, MDA, and water in brain tissue increased significantly compared with the saline control group. The pathological changes showed mixed brain edema. The results show that free radicals and lipid peroxidation are one of the important molecular pathological mechanisms of the formation and development of brain edema; in addition, H 2 O 2 solution can cause serious brain edema and injury, so clinical use should be avoided.
