目的:研究免疫八肽的纯化工艺。方法:以水-乙腈为二元梯度洗脱流动相,以0.1%三氟乙酸(TFA)为离子对,用RP-HPLC(反相高效液相色谱)对合成产物进行分析,探索主峰出峰时间和杂肽分离度,优化程序后将程序放大并制备纯化,用RP-HPLC分析纯肽产物并与标准品对照其色谱保留值,结果一致,结果:此程序下收集的纯肽产物是目标八肽,并且收集的3段肽液纯度全部大于95%,产率高,纯度高。结论:建立了适于大规模生产免疫八肽(AT-N)的纯化工艺。
Objective: To study the purification process of immune octapeptide. Methods: The mobile phase was eluted with binary gradient of water-acetonitrile and analyzed by RP-HPLC (reversed-phase high performance liquid chromatography) with 0.1% trifluoroacetic acid (TFA) as ion pair. Time and the degree of separation of the peptide. After optimizing the program, the program was enlarged and prepared and purified. The pure peptide product was analyzed by RP-HPLC and was compared with the standard value of the chromatographic retention value. The result was the same. The pure peptide product collected under this procedure was the target Octapeptide, and the purity of the collected 3-stage peptide liquid is more than 95%, with high yield and high purity. Conclusion: The purification process suitable for large-scale production of AT-N was established.
